I’d add though: my understanding is ion propulsion engines will allow us to get to significant fractions of a speed of light – 0.1c.

That would allow us to get to Alpha Centauri in about 44 years: well within a human lifetime.

All of the challenges around psychology and what not within the confines of a spaceship however would still apply.
